Inositol 1,4,5-trisphosphate receptor (IP3 R) functions as an intracellular Ca^2+ channel. It participates in the pathways of calcium-induced signal transduction and regulations of cellular activity through a change in intracellular Ca^2+ concentration when binding to its ligand, inositol 1,4,5-trisphosphate (IP3 ). In recent years, increasingly researches have shown that IP3 Rs played a vital role in the development of some cardiovascular disease, especially in heart failure, myocardial hypertrophy and atrial fibrillation. In this review, the recent advancement in structure and distribution of IP3 R, the mechanism it activates Ca^2+ channel and its relationship with cardiovascular disease are discussed.
