摘要
二胺氧化酶是具有高度活性的细胞内酶,能催化二胺类物质的分解.基于Lumlnol-Co^2+ -H2O2化学发光体系,将酶促反应与化学发光反应相耦合,并结合流动注射技术,通过检测H2O2的发光信号,间接测定二胺氧化酶的活性.该方法操作简单、快速、灵敏度高、重现性好.二胺氧化酶在1×10^-3 ~6×10^-2 U/L活性范围内与化学发光强度具有良好的线性关系,检出限为3.0×10^4 U/L,RSD为6.7%.
Diamine oxidase is a kind of intracellular enzyme which possesses high activity. It can catalyze diamines to produce hydrogen peroxide (H2O2). Based on the chemiluminescence system of Luminol-Co^2+ -H2O2, and combined with flow-injection technique, a chemiluminescence method for the determination of diamine oxidase was developed. The present method has following advantages:simplicity, rapid analysis, high reproducibility and sensitivity. The linear range is 1×10^-3 ~6×10^-2 U/L, the relative standard deviation (RSD) is 6.7% for 1× 10^-3 U/L of DAO with a detection limit of 3.0×10^4 U/L.
