摘要
目的:测定紫杉醇脂质体的药物包封率。方法:采用葡聚糖凝胶柱进行分离,HPLC法测定药物的含量。结果:用磷酸盐缓冲液作为洗脱液,在葡聚糖凝胶柱上能使包封药物与游离药物得到很好分离,结果表明混溶膜法制备的紫杉醇脂质体药物包封率最高。结论:该方法重现性好,柱子回收率高,是测定脂溶性药物包封率的首选方法。
Objective:To quantify the entrapment efficiency of paclitaxel liposomes. Methods: Paclitaxel was quantified by HPLC followed after a Sephadex G-50 chromatography. Results:The paclitaxel liposomes were separated well from free paclitaxel by the Sephadex G-50 chromatography with an eluate of PBS buffer. The optimal entrapment efficiency of paclitaxel liposomes was achieved using a mixed soluble membrane method. Conclusion:The reproducible and higher recovery Sephadex procedure was the best option in determination of entrapment efficiency of liposoluble drugs.
