摘要 目的:检测酸敏感离子通道(acid-sensing ion channels,ASICs)在唾液腺腺样囊性癌(ACC)中的表达情况,并探讨ASICs与ACC侵袭转移的关系。方法:采用免疫印迹和免疫组织化学方法,检测ASICs在培养的ACC-2细胞及腺样囊性癌组织中的表达。采用方差分析,利用OriginPro7.0软件进行统计学分析。结果:ACC-2细胞和腺样囊性癌组织中有ASICs蛋白的表达,而在正常组织中未见ASIC2a的表达;ASICs蛋白表达量经灰度值分析,正常腮腺组织为0.00012±0.00044,多形性腺瘤组织为0.00023±0.00049,腺样囊性癌组织为0.57±0.00045,差异有显著性意义,P<0.001。结论:腺样囊性癌中ASICs表达增多,可能是腺样囊性癌侵袭转移的组织病理学基础。 PURPOSE: To investigate the expression of acid-sensing ion channels (ASICs) in adenoid cystic carcinoma (ACC) of salivary glands, and evaluate the role of ASICs in the invasion of ACC. METHODS: Expression of ASICs in cultured ACC-2 cells and samples from normal salivary glands(NA) ,pleomorphic adenoma (PA) tissues or ACC tissues was detected by western blotting and immunohistochemistry. Statistical analysis was performed by one-way analysis of variance (ANOVA) with OriginPro software (Version 7.0). RESULTS: The enhanced expression of ASICs was observed in cultured ACC-2 cells and ACC tissues. Moreover, the enhanced ASIC2a expression was observed in the nuclei of ACC. (NA: 0.00012±0.00044, PA: 0.00023±0.00049,ACC: 0.57±0.00045, P〈0.001). CONCLUSION: The enhanced density of ASICs expression in ACC may contribute to the invasion of ACC.
