摘要
目的:观察18β-甘草次酸(18β-glycyrrhetinic acid,GA)对人胃癌细胞系BGC823细胞增殖的影响,并从细胞周期及细胞周期素依赖性激酶抑制蛋白(p16,p21,p27)方面探讨GA可能的作用机制。方法:以不同浓度的GA处理胃癌BGC823细胞后,用MTT法检测细胞增殖,以流式细胞仪检测细胞周期,采用Western blot方法检测胃癌细胞p16,p21,p27蛋白水平变化。结果:GA能抑制BGC823细胞增殖,且呈浓度依赖性。胃癌细胞经GA处理后,细胞周期发生G0/G1期阻滞。另外发现,胃癌细胞p21,p27的蛋白水平上调,但p16的蛋白水平没有明显变化。结论:GA可抑制人胃癌细胞增殖,其机制与G0/G1期阻滞、p21,p27表达上调有关。
Objective: To investigate the effects of 18β-glycyrrhetinic acid ( GA ) on proliferation, cell cycle and expression of cyclin-dependent kinase inhibitors (CKIs) of human gastric cancer cell line BGC823. Methods: After BGC823 cells treated with varible dose of GA, we detected the proliferation by MTT assay, and observed cell cycle by flow cytometer, studied the protein level of CKIs ( p16, p21, p27 ) by Western blot. Results: GA could inhibit the proliferation of BGC823, and led to G0/G1 phase block and up-regulate the level of protein of p21 as well as p27, but have no significant effect on the level of protein of p16. Conclusion.. GA can inhibit proliferation of human gastric cancer cell line BGC823 through arresting Go/G1 phase and up-regulating of expression of CKIs, especially p21 and p27.
