摘要
目的:探讨小分子活性肽apelin及其受体APJ在两肾一夹高血压病理进程中的变化及意义。方法:清洁级雄性SD大鼠20只,随机均分为对照组和模型组。测定平均颈动脉压(mCAP)、左室舒张末期压(LVEDP)以及左心室压力变化最大速率(LV±dp/dtmax),测定左心重与体重比值(LVM/BW)。以RT-PCR方法检测左心室组织apelin、APJ基因表达的变化。结果:①模型组大鼠mCAP较对照组高36.58%(P<0.05),LVEDP较对照组高333.8%(P<0.01),LVM/BW较对照组高20.24%(P<0.01),LV+dp/dtmax较对照组低85.35%(P<0.05)。两组大鼠LV-dp/dtmax差异无统计学意义。②模型组左心室组织apelinm RNA、APJ mRNA表达水平,比对照组分别高77.66%和119.00%(P均<0.05)。结论:高血压大鼠左心室apelin-APJ基因表达的上调在肾性高血压左心室肥厚的发生、发展过程中可能扮演重要角色。
Objective:To examine the expression changes of apelin and its receptor (APJ) genes in left ventricle tissues of two-kidney, one-clip(2K1C) rats. Methods: Twenty male Sprague-Dawley rats were randomly divided into normal control group and model group. The mean of carotid arterial pressure (reCAP), mean of left ventricular end diastolic pressure (LVEDP), the maximum rate of change in left ventricular pressure(LV ± dp/dtmax), and the weight ratio of left ventricular mass (left ventricle plus septum)to bodyweight (LVM/BW) were measured to estimate the model of 2K1C renal hypertension. The expression changes of apelin and APJ genes of left ventricle (LV) were detected with reverse transcription- polymerase chain reaction (RT-PCR). Results: (1)The mCAP, LVEDP and LVM/BW of 2K1C group were higher (36.58%, 333.8% and 20.24%,respectively) than those of normal control group (P〈0.05, P〈0.01, P〈0.01), but LV+dp/dtmax was lower 85.35%(P〈0.05). However LV-dp/dtmax was of no significant difference between two groups. (2)The expressions of apelin and APJ mRNA in left ventricle tissues of 2K1C rats were higher (77.66% and 119.00%,respectively) (all P〈0.05) than those of normal control group. Conclusion: The local increased expressions of apelin and APJ genes in left ventricle in renal hypertension rats may play an important role in the development of left ventricular hypertrophy.
