摘要
目的:观察骨髓间充质干细胞(BMSC)植入猪急性心肌梗死(AMI)区的转归及治疗作用。方法:选择10只体重35~45kg的家猪,经导管应用球囊封堵前降支动脉,建立心肌梗死模型,将存活的9只动物随机分为2组:(1)实验组5只,取骨髓分离BMSC体外培养扩增,建立模型2周后,通过心肌注射的方法将自体骨髓BMSC移植至AMI区;(2)对照组4只动物注射等量生理盐水。4周后比较2组的生存率、心功能变化、心肌功能性蛋白质表达的差异。结果:BMSC移植后,实验组动物心功能得到明显改善,梗死区室壁运动速度加快,局部室壁厚度加快,心肌移植细胞部位表达连接蛋白GATA-4等心肌功能性蛋白质。结论:骨髓BMSC经冠脉转运在心肌微环境下可向心肌细胞分化,并能改善心功能。
Objective: To study the therapeutic effect and differentiation of bone marrow mesenchymal cell (MSC) after transplantaion for pigs with acute myocardial infarction(AMI). Methods: Ten pigs, weight 35445 kg were used. The acute myocardial infarction (AMI) model was set up by occlution anterior descending coronary artery with balloon. Then the animals were divided into two groups randomly. In the therapeutic group, 80 ml marrow were aspirated and MSCs were islolated and cultured in vitro then were transplantated into AMI area. While 0. 9% sodium chloride were injected in the same way in control group. The implantated cells and cardiac special proteins of GATA 4 were examined by cellular immunochemical stain at 4 weeks. Echocardiography was performed to observe the changes of cardiac function before and after trassplantation. Results: Four weeks after cell transplantation, the beneficial effects on cardiac function were obvious. The hearts with transplanted cell had better I.V ejection fraction and infarction wall movement velocity than those of control hearts. All the hearts of MSCs transplanted pigs were found that new generated myocardiocytes existed aroud ischemic and infracted zone. Conclusion: Self transplantation of MSCs induced new generated cardiac cells in hearts of acute myocardial infarction pigs and therapeutic effectiveness in improving cardiac function.
