大鼠蛛网膜下腔出血不同时相脑组织C-fos表达实验研究 被引量:1

摘要 目的:探讨大鼠蛛网膜下腔出血不同时相的症状及行为表现与脑组织C-fos表达的关系及其生物学意义。方法:选用Wistar大鼠70只,建立大鼠蛛网膜下腔出血模型,随机分为蛛网膜下腔出血30min组、1h组、2h组、6h组、24h组、72h组、7d组,共7组,每组6只。对照组(生理盐水对照组)共7组,每组4只。取脑做连续切片后,采用HE染色,光镜下观察并进行定位。采用C-fos免疫组化染色,在显微镜下观察C-fos于不同时相在脑膜、脑皮质区及相关核团如尾壳核、苍白球、海马、背侧丘脑腹后外侧核、小细胞网状核、背侧旁巨细胞核的阳性细胞数。结果:对照组C-fos在尾壳核、苍白球、海马、背侧丘脑腹后外侧核、小细胞网状核、背侧旁巨细胞核表达极低;30min、1h、2h、6h组C-fos表达较高,与对照组比较差异均有统计学意义(P<0.001);24h、72h和7d组C-fos表达较低,但是与对照组比较差异也均有统计学意义(P<0.001)。结论:(1)大鼠蛛网膜下腔出血后不同时相C-fos在脑膜、脑皮质区及尾壳核、苍白球、海马、背侧丘脑腹后外侧核、小细胞网状核、背侧旁巨细胞核中都有表达且表达量随时相改变;(2)大鼠蛛网膜下腔出血后相关症状与不同时相C-fos在脑中表达之间存在着相关联系,具有规律性。 Objective: To observe C-fos expression in brain after rat subarachnoid hemorrhage (SAH). Methods: A classical animal model of rat SAH was established by imitation. 70 healthy rats were randomly divided into 7 groups each group was 6. After SAH for 30 min (groupⅠ), 1 h (group Ⅱ ), 2 h (group Ⅲ), 6 h (groupiv), 24 h (groupV), 72 h (groupⅥ), 7 days (groupⅦ) and seven blank groups. Rats were put to death after stimulus for 2 hours. Location under the microscope by using HE staining. The varying of expression of C-fos in different phases and different brain tissue including caudate nucleus, putamen, globus pallidus, hippocampus, nuclei ventrolaterales thalami, parvocellular reticular nucleus, and dorso lateral magnocellular nucleus were examined by immunohistochemical technique. Results: The expression of C-fos was the lowest in blank groups including caudate nucleus, putamen, globus pallidus hippoca, nuclei ventrolaterales thalami, parvocellular reticular nucleUs, dorso lateral magno cellular nucleus; the expression of C-fos were higher including caudate nucleus, putamen, globus pallidus, hippocampus, nuclei ventrolaterales thalami, parvocellular reticular nucleus, and dorso lateral magnocellular nucleus among groupI, group Ⅱand group Ⅲ group Ⅳ ; The expressions of C-fos were lower including caudate nucleus, putamen, globus pallidus, hippocamp, nuclei ventro laterales thalami (P 〈0.001), parvocellular reticular nucleus, dorso lateral magnocellular nucleus both group V , group VI and group Ⅶ (P 〈0.001). Conclusions: (1) Verification of the expressions of C-los in caudate nucleus, putamen, globus pallidus, hipp ocamp, nuclei ventrolaterales thalami, parv- ocellular reticular nucleus, dorso lateral magnocellular nucle us after rats subarachnoid hemorrhage are variation with different phases. (2) There are certain relation ships between subarachnoid hemorrhage and different phases.
