摘要
目的探讨幽门螺杆菌(Hp)感染对小鼠胃粘膜上皮细胞表达环氧合酶-2(cyclooxygenase-2,COX-2)的影响。方法32只Balb/c小鼠随机分为4组。第1组为对照组,口服液体培养基;第2、3、4组为实验组,分别等量口服Hp菌悬液。于末次灌胃后第10、10、24、32天将第1、2、3、4组小鼠处死。用快速尿素酶试验、Warthin-Starry银染和Hp培养作Hp感染诊断;用免疫组化比较各组COX-2表达量;用流式细胞仪检测小鼠胃粘膜上皮细胞增殖指数(PI)。结果对照组均无Hp感染,实验组均检测到Hp。第1组与第2、3、4组COX-2表达量差异有统计意义(P<0.01),第2组与第3、4组表达量差异也有统计学意义(P<0.01),但第3、4组间差异无统计学意义(P>0.05)。第1、2组间PI差异无统计学意义(P>0.05),其他各组间差异均有统计学意义(P<0.01)。结论在Hp感染初期小鼠胃粘膜上皮细胞就可能启动了由COX-2介导的细胞增殖机制,并参与了Hp相关胃癌发生、发展的过程。
Objective To explore the effect of Helicobacter pylori (Hp) on cyclooxygenase-2 (COX-2) expression in the gastric mucosa cells of the Balb/c mice. Methods A total of 32 Balb/c mice were divided into 4 groups. Group 1 was used as the control group and was given the corresponding buffer orally, while group 2, 3, 4 were inoculated with Hp. Group 1, 2, 3 and 4 were sacrificed after 10, 10, 24, and 32 days after the last inoculation. The infection of Hp in gastric mucosa was analyzed by use of rapid urease test, Warthin-Starry silver staining and Hp culture. COX-2 was detected immunohistochemically. Proliferation index (PI) was calculated by flow cytometry. Results Control group was Hp negative. Other three groups were all Hp positive. The expression levels of COX-2 were statistically different among the groups(P〈0.01 ) except group 3 and 4(P〉0.05 ). PI were statistically different among the groups(P〈 0.01 ) except group 1 and 2 (P 〉0.05). Conclusion At the initial stage of Hp infection, COX-2 may induce the growth of gastric mucosa cells. The expression of COX-2 may play a role in the development of the Hp associated gastric cancer.
