摘要
目的建立测定血浆中龙血素B浓度的反相高效液相色谱分析方法。方法用乙腈沉淀血浆蛋白,离心后取上清液进行色谱分析。分析柱为Eclipse XDB-C18,流动相为乙腈-0.1%磷酸(35∶65,V/V),流速为1.0 mL/min,检测波长280 nm。结果龙血素B在血浆检测浓度线性范围为0.3~3.6μg/mL(r=0.99475);在血浆中的相对回收率为97.4%,RSD=0.86%(n=4)。结论本方法准确可靠、操作简便,可用于龙血竭的临床药动学研究。
Objective To establish a biopharmaceutical analytical method for the detemainafion of loureirin B in human plasma by RP - HPLC. Methods The plasma proteins were precipitated with acetonitrile and the supematant liquid obtained from the serum centrifugate was subjected to chromatographic analysis. The analytical column was Eclipse XDB-C18, the mobile phase consisted of acetonitrile-0.1% phos- phoric acid(35 : 65, V/V) with a flow rate at 1 ml·min^ - 1, he detection wavelength was 280 nm. Results The linear range of loureirin B in plasma was 0.3 ~ 3.6 μg·mL^- 1 ( r = 0.99475 ). The relative recoveries of lottreirin B in human plasma were 97.4 %, RSD = 0.86 % ( n = 4). Conclusion This method is accurate,reliable and simple and it is suitable for the pharmacokinetic study of blood concentration of dragon' s blood.
