霍乱毒素或联合外周神经对再生视网膜节细胞c-Jun表达的研究

Effects of Choleratoxin or Combinated with Peripheral Nerve on c-Jun Expression in Regenerated Retinal Ganglion Cells

摘要 目的:观察成年金黄地鼠视神经近端微挤压断(MC)后,再生视网膜节细胞(RGCs)c-Jun的表达与再生的关系。方法:实验动物随机分为CTx组、SN+CTx组,每组5只。近端微挤压断视神经后,玻璃体内注射霍乱毒素(CTx)或联合植入小段坐骨神经分支(SN)。术后4W,用荧光金(FG)逆行标记和c-Jun免疫荧光组织化学双标法观察再生的RGCs和c-Jun的表达。结果:再生RGCs胞核内有c-Jun蛋白表达,CTx组表达c-Jun的再生RGCs为(317±61)个,约占其再生总数的89.9%;SN+CTx组c-Jun阳性再生的RGCs为(418±102)个,占再生总数的96.8%,两组相比差异有统计学意义(P<0.05)。结论:c-Jun表达可能与RGCs的再生有密切关系,霍乱毒素或联合外周神经可显著提高再生RGCs的c-Jun表达。 Objective:To investigate the expression of c - Jun in regenerating retinal ganglion cells (RGCs) after the proximate optic nerve micro - crushed ( MC ) in proximal end in adult hamsters. Methods : After the proximal ends of optic nerve (ON) had been micro -crushed, CTx were used for intravitreal injection. or combined with a 2 mm segment of sciatic nerve (SN) inserted intra - vitreouslly. The animals were divided into two groups:CTx group and SN + CTx group,5 animals in each group. 4 weeks later,the fluorogold (FG) retrograde labeling method and c - Jun immunofluorecence labeling method were used to estimate the expression of c -Jun in the regenerating RGCs. Results :The numbers of c -Jun positive regenerated RGCs was 317 + 61 per retina in CTx group ,which account for 89.9% of the total regenerating RGCs. CTx accompanied by SN can enhanced the regenerated c -Jun positive RGCs to 418 ± 102 per retina,which constituted 96.8% of the total regenerated RGCs, significantly higher than that in the CTx group (P 〈 0.05 ). Conclusions:Expression of c - Jun are closely related to the regeneration of RGCs. CTx or combined with SN can promote the c - Jun expression in regenerated RGCs.
