2-甲氧基雌二醇对C6脑胶质瘤细胞增殖与凋亡的作用

Effects of 2-methoxyestradiol on proliferation and apoptosis of C6 glioma cells

目的探讨2-甲氧基雌二醇对C6脑胶质瘤细胞增殖及凋亡的影响。方法以不同浓度的2-甲氧基雌二醇分别作用C6胶质瘤细胞不同时间,采用甲基噻唑蓝比色法(MTT)检测细胞活性,流式细胞仪检测细胞凋亡和细胞周期,并用光镜及电镜观察细胞形态学变化。结果经2-甲氧基雌二醇作用后,C6胶质瘤细胞活性受抑制,且呈时间依赖性,作用组与对照组之间的差异具有统计学意义(P<0.05)。此外,2-甲氧基雌二醇还可诱导C6胶质瘤细胞凋亡,凋亡率在作用组与对照组之间存在显著差异(P<0.05);细胞周期检测发现作用组G1及S期细胞减少,G2期细胞增多,与对照组存在显著差异(P<0.05)。结论2-甲氧基雌二醇可抑制C6胶质瘤细胞生长,诱导C6胶质瘤细胞凋亡。

Objective To investigate the effects of 2-methoxyestradiol on the growth and apoptosis of C6 glioma cells in vitro. Methods The C6 glioma cells were treated by different doses of 2-methoxyestradiol with different duration. They were observed by the microscope and electromicroscope. The viability was examined by methyl thiazolyl tetrazolium assay, and cell cycle or apoptosis was examined by flow cytometry. Results 2- methoxyestradiol caused a dose- and time-dependent inhibition on the C6 cell viability （P 〈 0. 05 ）. 2- methoxyestradiol could induce a dose and time-dependent apoptosis of C6 cells （ P 〈 0. 05）. After treating with 2- methoxyestradiol, the number of C6 cells in the G, and S phase decreased while that of G2 phase increased （P 〈0.05）. Condusion 2-methoxyestradiol can inhibit the proliferation and induce the apoptcsis of C6 ghoma cells.
