热休克蛋白对实验性肝纤维化中TGF-β_1的影响 被引量:2

摘要 【目的】观察热休克蛋白(HSP)高表达时在实验性肝纤维化大鼠中转化生长因子1(TGF-β1)表达情况,探讨肝纤维化发生过程中HSP对细胞因子TGF-β1的影响。【方法】用硫代乙酰胺诱导肝纤维化动物模型,采用免疫组化、计算机图像分析等技术,观察TGF-β1在正常对照组,肝纤维化模型组以及热休克反应后肝纤维化模型组中的表达和定位特点,检测HSP高表达时各组间TGF-β1的变化。【结果】TGF-β1在实验性肝纤维化发展时期,其表达增高差异有显著性(P<0.05)。热休克反应实验组中TGF-β1的表达可见明显降低(P<0.05)。【结论】TGF-β1在实验性肝纤维化发生发展中起着重要作用,而在HSP高表达时TGF-β1受到抑制。HSP可能通过抑制TGF-β1的产生而减缓肝纤维化发生。 [Objective]To study the expression of transforming growth factor-beta1 (TGF-β1 ) in experimental rat hepatic fibrosis and to discuss the possible effects of heat shock protein (HSP) on the expression of TGF-β1 in the development of liver fibrosis. [Methods]Liver fibrosis model rats were setup by thioacetamide. In all the tested animals , their liver tissues were obtained for immunohistochemistry staining. The expression degree of TGF-β1 and the changes of morphology in liver were measured by image analysis system. [Results] Fibrosis models were successfully constructed. The expression of TGF-β1 was increased in the processes of experimental hepatic fibrosis rats ( P〈0.05) . However, There was obvious decrease in the expression of TGF-β1 during the over-expression of HSP ( P 〈0.05). [Conclusion] TGF-β1 may play an important role in the development of hepatic fibrosis, while the over-expression of HSP could inhibit the expression of TGF-β1. HSP may delay development of liver fibrosis through inhibiting the expression of TGF-β1 .
