羊栖菜多糖对血管内皮细胞氧化损伤的保护和修复作用的实验研究 被引量:11

摘要 目的研究羊栖菜多糖(SFPS)对过氧化氢所致人脐静脉内皮细胞(HUVEC)损伤的保护及修复作用。方法噻唑蓝(MTT)法检测细胞活力,比色法测定细胞培养液中乳酸脱氢酶(LDH)活力、NO水平、超氧化物歧化酶(SOD)活性及细胞内丙二醛(MDA)含量。结果羊栖菜多糖可剂量依赖性地抑制过氧化氢所致的细胞活力降低,阻止LDH外漏,增加NO释放,并减少MDA生成,提高SOD活性。结论羊栖菜多糖对过氧化氢所致HUVEC损伤具有明显的保护及修复作用。 OBJECTIVE To study protective and repairing effects of sargassum fusiforme polysaccharides (SFPS) on human umbilical vein endothelial cells(HUVEC) injured by hydrogen peroxide in vitro. METHODS The activity of HUVEC was monitored by MTT assay. The activity of LDH, SOD and NO level in culture medium was measured by colorimetric, so was MDA content of HUVEC. RESULTS SFPS inhibited the activity of HUVEC injured by hydrogen peroxide, prevented the release of LDH and increased the level of NO in a dose-dependent manner. Furthermore, SFPS reduced MDA content and enhanced the activity of SOD. CONCLUSION SFPS has obvious protective and repairing effects on HUVEC injured by hydrogen peroxide.
