摘要
为探讨海南原生兰花花粉萌发所需的培养基成分,测定其生活力提供技术和方法,以海南原生墨兰为材料,采用不同的液体培养基进行花粉萌发试验。试验结果表明:海南原生墨兰花粉在所设计的液体培养基上都易萌发且萌发花粉管长。除对照外,7个处理萌发率高达78%以上。在含10%蔗糖,20mg/L硼酸和5mg/LCaCl_2的液体培养基上培养的萌发率为83.99%,效果较好,本着简便原则,可应用于海南原生墨兰花粉生活力的鉴定。
For discussing on the optimizing composition of original orchids pollen germination mediums in Hainan Province, and testing their pollen viability, several liquid mediums were designed for pollen getmination experimentation of Cymbidium sinense which was original in Hainan Province. The experimentation indicated that Cymbidium sinense pollen original in Hainan Province readily germinated in artificial liquid mediums in the experiments with long pollen tubes. The average germination percentages of seven treatments up to 78% except the contrast treatment. It was 83.99% on liquid medium containing 10% sucrose, 20mg/L boric acid and 5 mg/L calcium chloride with optimizing result. For simple principle, it will be used for identifying Hainan Cymbidium sinense pollen viability in future.
