摘要
目的:观察电针对慢性疲劳大鼠模型下丘脑—垂体—肾上腺轴(HPA-axis)激素水平的影响,探讨电针(EA)治疗慢性疲劳综合征(CFS)的作用机理。方法:设正常组、模型组、电针组,模型组与电针组采用强制性冷水游泳的方法制作CFS大鼠模型,电针组在造模的同时在百会、足三里穴给予电针治疗。通过行为学检测对模型进行判定;采用酶联免疫吸附测定(ELISA)法检测血清促肾上腺皮质激素(ACTH)和皮质醇(CORT)的含量;采用放射免疫分析法测定大鼠血清下丘脑促肾上腺皮质激素释放激素(CRH)含量。结果:①电针对慢性疲劳大鼠行为学的影响:模型组大鼠行为出现改变,产生了心理和躯体的疲劳状态,基本符合或类似于CFS的临床表现。电针对疲劳大鼠模型的心理和躯体疲劳状态有显著良性调节作用。②电针对慢性疲劳大鼠HPA轴的影响:模型组大鼠HPA轴功能出现异常,血清ACTH、CORT和CRH含量明显上升;电针能明显降低血清ACTH、CORT和CRH的浓度,使HPA轴功能恢复正常。结论:①CFS与应激关系密切,采用强制性冷水游泳制作的慢性疲劳大鼠模型与CFS的发病机制和临床表现基本相符。②电针治疗CFS的机制可能与其对HPA轴功能的调节有关。电针可调节血清ACTH、CORT和CRH的浓度,在3个层次对HPA轴的激素进行调节,以纠正CFS异常的HPA轴功能,达到治疗CFS的目的。
Objective: By observation in the regulative effectiveness of electro -acupuncture (EA) on hypothalamic - pituitary - adrenal axis ( HPA - axis) for Chronic Fatigue Syndrome (CFS) model rats, studying the mechanism of CFS with treatment of EA in the point of the neuroendocrine system. Methods : Experimental rats were randomly divided into normal control group, model group, electro - acupuncture group, Model and EA groups were adopted forced cold water swimming stress to make fatigue model, while EA group adopted electro - acupuncture treatment at the same time. EA group was punctured in Baihui( DU20), Zusanli(ST36). Evaluation of the model was processed according to the behavior changes of the rats. Adrenocorticotrophic hormone(ACTH) , cortisol (CORT) in serum were measured by enzyme linked immunosorbent assay (ELISA). Corticotropin releasing hormone (CRH) in hypothalamic were measured by radioimmunoassay. Results : ①The effects of EA on ethological for CFS model rats : The model rats behavior changed, both of physical fatigue and mental fatigue were observed. They reflect that the rat model induced by complex stressors was basically successful in simulating the pathogenesis and clinical manifestations of CFS. EA showed regulative effectiveness in physical fatigue and mental fatigue of the CFS model rats.②The regulative effects of EA on HPA - axis for CFS model rats: Disorder of HPA -axis were detected in the CFS model rats, ACTH ,CORT and CRH in the serum obviously raised, EA could reduce the level of ACTH ,CORT and CRH in in serum significantly, make the HPA axis function to normal state. Conclusion: ①Stress is closely related factors which induce to CFS. CFS model rats made by forced code water swimming as complex stress factors is a successful copy of the CFS as confirmed by observation of the clinical symp- toms. ②EA could obviously improve to the fatigue symptom of CFS model rats, both in physical fatigue and mental fatigue. EA could regulate the function of HPA - axis, which were possible the mechanism of electro - acupuncture to achieve the therapeutic effects. EA could modulate CRH, ACTH and CORT , and accordingly regulate HPA - axis from three levels. CFS could be treated by EA effectively.
