降铅Ⅰ号对低水平铅暴露仔鼠血铅及海马一氧化氮合酶的影响

摘要 目的:观察降铅I号对低水平铅暴露仔鼠的血铅及学习记忆能力的影响并探讨其作用机制。方法:选用Wistar大鼠仔鼠50只,随机分成5组,每组10只,为模型组、EDTA对照组和降铅Ⅰ号低剂量、中剂量和高剂量治疗组。喂0.2%醋酸铅的去离子水造模后,每组均随机选取2只仔鼠,检测血铅及海马一氧化氮合酶(NOS)活性的变化。其余仔鼠给药30天后,再检测以上指标。所有大鼠进行Y迷宫实验观察降铅Ⅰ号对铅中毒大鼠学习记忆能力的影响。结果:降铅Ⅰ号治疗30天后,各治疗组血铅水平都较模型组下降,海马NOS活性除低剂量组外,其余各组都有升高,差异有显著性(P<0.05)。Y迷宫实验显示,降铅I号治疗组的记忆保持率高于模型组,疗效以中、高剂量组为优,与低剂量组比较,差异也有显著性(P<0.05)。结论:降铅Ⅰ号有较强的驱铅及改善学习记忆能力的作用。升高海马NOS活性可能是其改善学习记忆能力,减轻铅神经毒性的作用机制之一。 Obiective: To investigate the effects of JiangQian Ⅰ on blood-lead and ability of learning and memory in rats exposed to low level lead and explore its possible mechanism. Methods: 50 Wistar rats were divided into five groups randomly,which named model group,EDTA group,JiangQian Ⅰ low dose group,middle dose group and high dose group. Model rats which poisoning of lead were prepared by drinking water with lead acetate. Then ten rats were choused randomly from each group to measure the lead content in blood and nitric oxide synthase (NOS) activity in the hippocampus. They were treated for 30 days and the same target were measured too. All rats were taken Y maze to test the ability of leaning and memory which indicated the lead-induced injuries to nervous. Result: After 30 days treatment with JiangQian Ⅰ, The lead content in blood of each treatment group was significantly lower than the model group and the NOS activity in hippocampus was higher than the model group(except JiangQian Ⅰ low dose group)( P 〈0.05) ,Y maze test indicated that the retain percent of learning and memory of three groups treated with JiangQian Ⅰ was obviously increased than the model group. Futhermore,the effect of two groups treating with JiangQian Ⅰ middle dose or high dose is better than the group treating with JiangQian Ⅰ low dose( P 〈0.05). Conclusion. JiangQian Ⅰ has curative effect to rats poisoning of lead and can'amend the impairment of learning and memory. JiangQian I can increase the NOS activity in hippocampus, which may be the mechanism of it.
