密蒙花提取物对去势导致干眼症白兔泪腺细胞凋亡的影响 被引量:50

摘要 目的观察密蒙花提取物对去势导致的干眼症兔基础泪液分泌量、泪膜稳定性和凋亡相关基因蛋白Fas、FasL表达的影响,探讨密蒙花提取物抗性激素水平失调导致的干眼症的作用机制。方法将60只日本大耳白兔(雌雄各半)随机分为雄、雌兔空白组(A1、A2),雄、雌兔模型组(B1、B2),雄、雌兔1倍剂量密蒙花提取物治疗组(C1、C2),雄、雌兔2倍剂量密蒙花提取物治疗组(D1、D2),雄、雌兔染料木黄酮治疗组(E1、E2),每组6只。对B、C、D、E组行去势术建立动物模型,对C、D、E组分别以密蒙花提取物1倍剂量和2倍剂量以及染料木黄酮连续灌胃1月。对全部实验兔行SchirmerI试验、测量泪膜破裂时间,用免疫组织化学的方法对兔泪腺组织中凋亡相关基因蛋白Fas、FasL进行检测。结果C、D、E组SchirmerI试验测量值明显高于B组(P<0.01)、泪膜破裂时间明显长于B组(P<0.01),泪腺导管及腺泡上皮细胞中,Fas、FasL阳性表达的细胞数均明显低于B组(P<0.01)。以上检测结果C、D组优于E组(P<0.05)。结论密蒙花提取物中主要成分为黄酮类物质,可显著抑制性激素水平降低后兔干眼症的发生。其作用机制可能与黄酮类物质为植物雌激素的一种有关,可调节体内性激素水平,从而可调节和抑制泪腺细胞凋亡。密蒙花提取物可能成为治疗干眼症的新途径。 OBJECTIVE To assess the roles of extract of Buddleja officinalis in basic tears secretory volume, tear film stability, expression of Fas, FasL in lacrimal gland of castrated rabbits with dry eye, and to discuss the therapeutic effects of extract of Buddleja officinalis on dry eye caused by gonadal hormones level imbalance.METHODS Sixty rabbits (male and female were respectively half of all)was divided averagely into normal group (A 1 and A2 ), model group( B 1 and B2) ,therapy group with single- dose of Buddleja officinalis extract( C1 and C2), therapy group with double- dose of Buddleja officinalis extract( D1 and D2)and therapy group with genistein( E1 and E2). The dry eye model was established with ORX and OVX on Group B, C, D, E. Group C, D, E were gastrically perfused with single - dose, double - does of Buddleja officinalis extract and genistein respectively for 30 days. All rabbits were detected with SIT and BUT. Fas and FasL were detected on immunohistochemistry. RESUTS The SIT value of group C, D, E is significantly higher than that of group B( P 〈 0.01 )and the BUT value of group C, D, E was significantly longer than that of group B ( P 〈 0.01 ). The expression of Fas, FasL in acinar cell and glandular tube cell of group C, D, E were significantly higher than that of group B( P 〈 0.01).Further,any result of group C,D was better than that of group E( P 〈0.05).CONCLUSIONS The main components of extract of Buddleja offlcinalis are flavonoids, which are phytoestrogens. Phytoestrogen displays estrogen- like activity, so it can adjust endogenous gonadal hormone level. Extract of Buddleja officinalis may be an effective therapeutic to this kind of dry eye syndrome because of its active mechanism. Thus, further research is warranted.
