新陆早19号胚珠培养体系的建立 被引量:4

摘要 在Beasley和Ting方法的基础上,建立了新疆陆地棉品种新陆早19号棉花胚珠离体培养纤维体系。在不继代的条件下培养25 d,在培养期内,测定了离体胚珠培养纤维生长长度,并首次利用数字化的图像和图像处理软件确定了离体胚珠及纤维生长轮廓所形成的面积。测定结果表明,在离体培养发育早期(前8 d),纤维发育迅速,其伸长长度绝对值相对较高,第20 d时已接近纤维的最终长度。但生长10 d以内的纤维强度很低,影响长度测量的准确度。培养前15 d离体胚珠生长的轮廓面积增长较快,15 d以后增长变缓,第20 d接近轮廓最大面积。 Based on the method developed by Beasley and Ting, an ovule culture system of Xinluzao No. 19 upland cotton was established. The ovules were cultured for 25 d under the condition of unchanging the culture media. During the culture, the elongated fiber length of cultured ovule was measured and the formatted area by the outline of cultured ovule and fibers was determined. The results showed that the fibers were elongated faster and the absolute elongated lengths were longer during early days of fiber development. However, the strength of the fiber developed in 10 d was very low. It affected the accuracy of the measurement. At 20 d, the elongated fiber almost reached the final - length. The formatted area by the outline of cultured ovule increased faster in 1 5 d before culture , and then slowly increased gradually after 1 5 d . The area reached the maximmn - area at 20 d.
