摘要
介绍了一种用反相高效液相色谱定量分析植物半乳糖脂(MGDG和DGDG)含量的方法.该方法采用岛津Shim-packCLC-ODS(150mm×6mm)柱,以体积比为9550.2的甲醇-水-乙酸溶剂为流动相,流速2mL/min,检测波长为205nm,灵敏度为0.032AUFS.在10~3500μg/mL浓度范围内成良好线性关系.MGDG和DGDG的回收率分别为96.7%和95.3%,时间保留值分别为16.625min和12.623min.方法简单快速,适于植物体内全脂中半乳糖脂含量的测定,为研究植物体内半乳糖脂的分布、生物学功能和代谢变化提供了有效的分析手段.
A quantitative analysis method of monogalactosyldiglyceride (MGDG) and digalactosyldiglyceride( DGDG) by reversed phase high performance liquid chromatography is established with methanol water acetic acid (95∶5∶0.2,v/v) as the mobile phase on Shimpack CLC ODS column with sensitivity of 0.032 AUFS.The flow rate is 2 mL/min.The reversed phase HPLC analysis at 205 nm is appropriate to determine the galactolipid content in total lipids of plant. Linearity of the measurement is in the range of 10 ̄3 500 μg/mL.The recovery rate of MGDG and DGDG is 96.7% and 95.3% respectively.Retention time of MGDG is 16.625 minutes and DGDG is 12.623 minutes. The method is simple, efficient in performance and can be applied to the study of distribution and function and metabolism of galactolipids in plant tissue.
