两性霉素B粗制品的结晶提纯工艺研究

摘要 以两性霉素B粗制品为原料,以产品中未知杂质含量、效价及质量收率为考察对象,探索了两性霉素B结晶的操作条件。实验发现,以20%N,N-二甲基甲酰胺-乙醇(75%)水溶液为溶剂体系、pH为5~6、75%乙醇水溶液加入量为3~4倍滤液体积时可得到杂质含量低、效价和质量收率较高的两性霉素B产品。 Taking the content of unknown-impurity in product, its potency and yield as the object for investigation, the crystallization process of amphotericin B was studied. The experiment results showed that the total content of the impurities in amphotericin B produced by this process could be decreased, and its potency and yield was higher when the aqueous solution composed of 20% DMF-ethanol (75%) was used as solvent system and pH value was at the range of 5 -6 and the volume of aqueous 75% ethanol solution in 3 -4 times as much as that of filtrate was added.
