摘要 通过研究,建立了适合于水稻根蛋白质双向电泳分析用的磷酸缓冲液抽提、TCA沉淀法浓缩蛋白质的新方法.本法用于水稻胚、叶片的蛋白质样品制备,也能获得稳定、清晰的双向电泳图谱.经考马斯亮蓝R250染色,根、叶和胚的清晰可分辨的蛋白质斑点数分别约为350、350和400个,用银染色,斑点数分别为600、600和700个.三者蛋白质的等电点总在3.0~9.0之间,大多在5.0~8.0之间,分子量在15~170kD之间。 A method suitable for analysis of root proteins using two dimensional electrophoresis was established in this study. It involved the extraction and precipitation of proteins using phosphate buffer and TCA. Stable and clear two dimensional electrophoresis map can be obtained when applied to proteins extracted from rice embryos and leaves using this method.The number of protein spots visualized with Coomassie Brilliant Blue staining is 350,350 and 400 in root, leaf and embryo respectively, and 600, 600 and 700 respectively, with silver staining.Their isoelectric points were found to lie between 3.0 and 9.0,most of them were in the range of 5.0~8.0. The molecular weights were between 15 to 170 kD, most of them lying in the range of 25~90 kD.
