摘要
论述了真菌脂肪酶在PEG/硫酸铵双水相体系中的分配行为,并研究了PEG分子量、PEG浓度、(NH4)2SO4浓度、NaCl等因素对脂肪酶和总蛋白分配系数、酶活回收率、相体积比的影响,并得到较好的分离条件.实际表明用14%~16%(W/W)PEG1000和12%~14%(W/W)(NH4)2SO4、NaCl为零所组成的双水相体系,在室温下对脂肪酶的提取率可达71%,分配系数可达1.7,提纯倍数为1.5.
The paper discussed the distribution of fungus lipase in the PEG/(NH 4) 2SO 4 two phase system. In the paper the dependence of partition coefficient of lipase and total protein,and the volume ratio of the top phase to the bottom phase on the average molecular weight of PEG,the concentration of PEG, the type and concention of salts included in the system was studied emphatically, and the appropriate separation conditions were determined. The suitable extraction conditions were 14~16%(W/W)PEG1000,14%~16% PEG 1 000, 12%~14%(W/W)(NH 4) 2SO 4,PH 8.5,without NaCl. It was shown that the partition coefficient of K e of 1.7,the yield of 71%,and 1. 5 folds increase of purity can be obtained.
