摘要
目的探讨细胞凋亡及Fas/FasL mRNA的表达在肺纤维化过程中的作用。方法通过博来霉素复制大鼠肺纤维化模型,同时设立正常对照组,采用TUNEL法、RT-PCR法检测肺组织细胞凋亡以及Fas/FasL mR-NA表达。结果肺纤维化模型复制成功。模型组肺组织细胞凋亡上调可持续至第28d,于第14d前后达高峰。Fas/FasL mRNA表达与细胞凋亡呈同步改变。结论细胞凋亡及Fas/FasL mRNA表达的上调可能是博来霉素致大鼠肺纤维化发生、发展的重要原因。
Objective To investigate the significance of cell apoptosis and FAS/FAS-L mRNA in the lung tissue of rats with pulmonary fibrosis. Methods The pulmonary fibrosis model in rat was established by bleomycin. TUNEL and RT-PCR were used to observe the cell apoptosis of rat's lung tissue and mRNA expression. Results The model was reproduced successfully. Compared with the control group, in model group the cell apoptosis was kept up-regulating from the 1st day to the 28th day and the peak appeared around the 14th day. The situation of the expression of FAS/FAS-L mRNA was the same as that of cell apoptosis. Conclusion The up regulation of cell apoptosis and the expression of FAS/FAS-L mRNA may play an important role in the development of pulmonary fibrosis.
