摘要
目的:复制家兔胆固醇结石模型,检测模型家兔肝组织固醇携带蛋白2(SCP2)mRNA的表达.方法:健康北京大耳白家兔16只,随机分为2组,每组8只:对照组喂普通饮食,模型组喂高胆固醇饮食(含12g/kg胆固醇).4wk时剖杀动物,采用半定量RT-PCR检测SCP2mRNA在家兔肝组织的表达.结果:对照组无结石形成;模型组成石率达87.5%.与对照组相比,模型组动物肝组织SCP2mRNA表达升高(0.79±0.17vs0.44±0.07,P<0.05).结论:高胆固醇膳食致家兔胆囊结石形成时肝组织SCP2mRNA表达升高.
AIM: To investigate the expression of sterol carrier protein 2 (SCP2) mRNA in liver tissues of rabbit model with cholesterol gallstone. METHODS: Beijing white rabbits (n = 16) were randomized into 2 groups: control group and model group (n = 8 per group), cholesterol gallstone was induced in model group by high cholesterol diet (containing 12 g/kg cholesterol). Liver tissues were harvested from these rabbits after 4- week feeding and SCP2 mRNA expression was detected by using semi-quantitative RT-PCR. RESULTS: After 4-week feeding, control group had no gallstone formation and 87. 5% of model group had formed stones in gallbladder. The expression of SCP2 mRNA was increased in model group as compared with control group (0.79±0.17 vs 0.44±0.07, P〈0.05). CONCLUSION: Expression of SCP2 mRNA in liver tissue is increased in high-cholesterol diet induced gallstone models.
