摘要
目的探讨大鼠供肝冷保存过程中腔静脉灌洗液中肿瘤坏死因子(TNF-a)水平随时间的变化及其与供肝质量的关系。方法建立大鼠肝移植的供肝切取的动物模型,切取供肝后0℃~4℃冷保存。将实验动物分为7组,每组动物6只,分别在0、4、8、12、16、20、24小时取冷保存时腔静脉灌洗液标本lml,测肝功能和TNF-a浓度。供肝做石蜡切片病理检查。结果在热缺血时间相同的情况下,大鼠供肝腔静脉灌洗液中肝功能水平和TNF-a水平随冷保存时间延长而增加。大鼠供肝腔静脉灌洗液中TNF—a水平与供肝质量相关。结论肿瘤坏死因子在肝脏缺血再灌注损伤中起重要作用,是介导损伤机制的重要细胞因子之一,其有作为评价供肝质量的一种指标的潜在价值。
Objective To investigate the difference of the level of TNF-a in the graft caval effluent after flushing of the liver with the cold preservation time changed and the relationship of the level of TNF-a and the graft quality. Method The animal model was liver harvest of rat liver transplantation, and preserved the liver in 0-4℃ UW fluid and the experimental animals were divided into seven groups according to the time. The collected samples were examined for liver function tests and TNF-a concentration. Pathology examination was done in the donor liver. Results With the same warm ischemic time, the TNF-a concentration increased with the cold preservation time. We found obvious morphological changes through the pathology examination in the donor liver. Conclusion TNF-a plays an important roles in liver ischemic repurfusion injury, and is a main cytokine to mediate the injuries. Through this experiment, we believe that the TNF -a concentration in the graft caval effluent is a potential index in evaluating liver donor quality.
