摘要 研究了脂肪酶拆分外消旋6-羟基-8-氯辛酸乙酯。从10种脂肪酶中筛选出了脂肪酶Lipase PS-D,该酶能够有效拆分外消旋6-羟基-8-氯辛酸乙酯,并对反应条件进行了优化,确定了该酶的最适反应条件:温度30℃,pH=7.0,摇床转速170 r/min,确定了有效表面活性剂为乳化剂OP,在此优化条件下反应1 h,得到(R)-6-羟基-8-氯辛酸乙酯的对映体过量值ee为93.1%,底物摩尔转化率为54.3%,总收率为91.4%。该研究为(R)-硫辛酸的制备提供了一条可行途径。 For lipase catalyzed resolution of racemic ethyl 6-hydroxy-8-chlorocaprylate, lipase PS - D was selected from ten enzymes because of its high catalytic activity and enantioselectivity. Under the optimal conditions that the reaction temperature, buffer pH and shaking rate were 30 ℃, pH 7.0 and 170 r/min respectively, a substrate enantionmeric excess (ee) of 93.1% was achieved with a yield of 91.4% after 1 h reaction time. This work provides to a feasible approach to the enzymatic production of (R) -lipoic acid.
