摘要
目的:观察磷脂酰胆碱注射液Lipostabil的局部注射溶脂作用。方法:实验于2006-08/10在南方医科大学动物实验中心完成。①实验分组:成年广西巴马小型猪12头,体质量50~60kg,将12头猪分别按序编号,随机分为实验组和阴性对照组,每组各6头。随机选择动物左侧或右侧腰背部、侧腹部、大腿局部范围为药物注射区,对侧为空白对照。②实验方法:在实验组猪皮下脂肪组织、真皮组织与肌肉组织内分别局部注射磷脂酰胆碱注射液Lipostabil0.5mL,共5次,每周1次。阴性对照组猪给予等量生理盐水注射。③实验评估:大体观察皮肤注射区表面红肿情况、组织病理学检查注射区局部组织变化及毒副作用,B超分别测量注射区及其对侧空白对照区的脂肪层厚度。结果:纳入成年广西巴马小型猪12头,均进入结果分析,未发现明显的全身毒副作用。①皮肤脂肪层注射区表面红肿情况:实验组猪皮肤注射区在药物注射后,皮肤红肿严重,有局部坏死发生;阴性对照组皮肤反应轻微。②磷脂酰胆碱注射液Lipostabil对实验组猪的皮下脂肪组织结构产生了明显的破坏作用,阴性对照组未见明显破坏。③B超测量注射区及其对侧空白对照区脂肪层厚度:与对侧空白对照区相比,实验组脂肪层厚度有不同程度的变薄[(0.444±0.284),(0.542±0.227)cm,P<0.01],阴性对照组无明显差异[(0.539±0.036),(0.538±0.036)cm,P>0.05]。磷脂酰胆碱注射液Lipostabil注射区的肌肉组织也有变性坏死。结论:磷脂酰胆碱注射液Lipostabil局部注射溶脂具有一定的作用,但其对组织的破坏不具有特异性。在进行进一步的研究前,不推荐进入临床应用于局部注射溶脂。
AIM: To evaluate the fat-dissolving effect of local injection of phosphatidylcholine Lipostabil.
METHODS: The experiment was carried out at the Animal Experimental Center of Southern Medical University from August to October 2006. Twelve Guangxi Bama miniature pigs of 50-60 kg ware numbered and divided into experiment group and control group with 6 animals in each group. The animals' left or right back, lateral region of abdomen, and thigh were randomly selected as injection regions, and the contralateral location served as blank control. The pig subcutaneous fatty tissue, epidermis, and muscular tissue of the experiment group were locally injected with 0.5 mL phosphatidylcholine Lipostabil for 5 times, once a week, While the pigs of the control group were given 9 g/L saline solution. The red swelling of the injection locations was grossly observed, and the tissue changes and the toxicity and side effects were detected by histopathology. Meanwhile, the thickness of fat layer of the injection locations and the contralateral locations was measured by ultrasonography.
RESULTS: All 12 experimental pigs entered the result analysis, and no obvious toxicity or side effects on the whole body was found. (1) In the injection locations of skin, severe red swelling even local necrosis were found in the experiment group, while the previous symptoms ware slight in the control group. (2) The phosphatidylcholine Lipostabil obviously destroyed the fat tissue of the experiment group, which was not obvious in the control group. (3)Compared with the contralateral locations, the thickness of fat layer of the injection locations was thinner in the experiment group [(0.444±0.284), (0.542±0.227) cm, P 〈 0.01], while the difference was not remarkable in the negative control group (0.539±0.036), (0.538±0.036) cm, P 〉 0.05]. Moreover, the injection locations of muscles were also found degenerated necrosis.
CONCLUSION: Local injection of phosphatidylcholine Lipostabil has a fat-dissolving effect to some extent, but its damage to tissues is not specific, so it needs further research before being used clinically for fat dissolving.
