间充质干细胞生物学特征及体外向血管内皮细胞的诱导分化 被引量:8

摘要 目的:间充质干细胞具有跨胚层分化等特性,因其体外分离培养可分化为血管内皮细胞,成为血管组织工程学的研究热点。本文对近期国内外间充质干细胞生物学特性和向血管内皮细胞分化的研究作一综述。资料来源:应用计算机检索Medline数据库2000-01/2007-02间充质干细胞生物学特性和向血管内皮细胞诱导分化的文章,检索词“endothelial,mesenchymal stem cells,differentiation”,限定文章语言种类为English。同时计算机检索中国期刊全文数据库、万方数据库2000-01/2007-02期间的相关文章,检索词“血管内皮细胞,间充质干细胞,分化”,限定文章语言种类为中文。资料选择:对资料进行初审,选取符合要求的文章查找全文。纳入标准:①有关间充质干细胞生物学性状的研究。②有关间充质干细胞诱导分化为内皮细胞方面的研究。排除标准:重复研究。资料提炼:共收集到95篇有关间充质干细胞生物学特性及向血管内皮细胞诱导分化的文章,排除重复的同一研究,28篇文章符合要求。资料综合:①国内间充质干细胞生物学特性及向血管内皮细胞定向诱导分化的研究:利用间充质干细胞向内皮细胞分化的特性在相关实验研究方面已取得一定的进展。②国外间充质干细胞生物学特性及向血管内皮细胞定向诱导分化的研究:研究的重点在间充质干细胞诱导分化为内皮细胞的机制,并在动物实验方面取得了良好效果。③间充质干细胞生物学特性及向血管内皮细胞定向诱导分化的研究展望:利用间充质干细胞生物学特性和血管组织工程学技术等方法为临床血管移植提供新策略。结论:通过间充质干细胞的分离扩增技术并添加内皮细胞诱导剂,在体外定向诱导出内皮细胞,这将为血管组织工程学提供广阔的应用前景。 OBJECTIVE: Mesenchymal stem coils (MSCs) have many characteristics such as transdifferentiation. Many researchers have confirmed that MSCs could differentiate into vascular endothelial cells in vitro, which becomes the hot spot of vessel tissue engineering. In this paper, the researches at home and abroad about the biological characteristics of MSCs and their differentiation into vascular endothelial cells in the recent years are reviewed. DATA SOURCES: A computer-based online search of Medline database was undertaken to identify the articles about the biological features of MSCs and differentiation into vascular endothelial cells published between January 2000 and February 2007 with the keywords of "endothelial, mesenchymal stem cogs, differentiation". Meanwhile, Chinese Journal Full-text Database and Wanfang database wore searched to identify the related articles published from January 2000 to February 2007 with the same key words in Chinese. STUDY SELECTION: The data were selected firstly, and only those articles that contained (1) researches of the biological characteristics of MSCs, and (2)researches of differentiation into endothelial cells were included. And the duplicated researches were excluded. DATA EXTRACTION: Totally 95 related articles wore collected, and 28 articles were finally selected after the duplicated or similar researches were excluded. DATA SYNTHESIS (1)Among the researches about the biological characteristics of MSCs and differentiation into endothelial cells in China, the related experimental researches about the inducing differentiation into endothelial cells have made a progress. (2)While the researches at abroad have focused on the mechanism of the induction and differentiation of MSCs into endothelial cells, and have obtained good results in animal experiments. (3)The biological characteristics of MSCs and technology of vessel tissue engineering could provide new strategy for vessel transplantation in clinic. CONCLUSION: After the separation and amplification, the MSCs could be induced to differentiate into endothelial cells by adding the revulsant of endothelial cells, and this may develop an extensive application prospect for vessel tissue engineering.
