昆明系和Balb/C系小鼠用于显性致死试验的比较研究 被引量:4

摘要 本研究选用昆明系和Balb/C系小鼠同时进行了显性致死试验的对比研究。试验一选用封闭群昆明系9~10周龄的雄性小鼠78只,随机分成6组,其中5个剂量组分别灌服160、53、16mg/kg体重的复方硝基酚钠和15、30mg/kg体重的环磷酰胺,另一组为空白对照组。受试物组每天给药一次,连续5d后,按1∶2的雄雌比例与正常的9~10周龄封闭群昆明系雌鼠进行交配,每周交配一批,连续交配8批。试验二除选用近交系Balb/C雄鼠取代昆明系雄鼠外,其它条件与方法同试验一。试验结果表明,各批雌鼠各组之间的受孕率、着床数均无明显差异;两个试验中15、30mg/kg环磷酰胺组的第1、第2和/或第3批的活胎数明显低于其它各组,死胎数、有一个以上死胎的孕鼠率及显性致死率明显高于其它各组,显性致死试验结果为阳性;而不同剂量的复方硝基酚钠组的各项指标与空白对照组比较未见明显差异。本研究中雌雄试验动物选用同一个品系的昆明系小鼠进行显性致死试验的结果与选用Balb/C系雄鼠与昆明系雌鼠进行杂交的结果比较无明显差异。 This report deals with the dominant lethal test in two trials of Kunming and Balb/C strain mice.In the 1st trial,78 male mice of closed Kunming strain were selected and divided into 6 groups (13 per group):3 groups(160,53,16mg/kg b w )of composed sodium nitrophenolates(CO NP Na),2 groups(30,15mg/kg b w )of cyclophosphamide and 1 control group After administering tested medicine for 5 days by oral,the male mice of each group mated with the Kunming female mice(1∶2),the female mice were exchanged each week,the male mice mated for 8 batches The method of the 2nd trial was the same as the 1st trial except substituting Balb/C strain male mice for Kunming strain male mice The results show that there were not significant differences on fertility and No of implants of each batch female mice In the 15 and 30mg/kg cyclophosphamide groups of two trials,No of living fetuses was significantly decreased and No of dead fetuses,ratio of pregnant mice which had more than one dead fetuses,rate of dominant lethal were significantly increased in the 1st,2nd and/or 3rd batches female mice The dominant lethality of cyclophosphamide was positive Comparing with the control group,the results of each group of CO NP Na in two trails were not significantly different In conclusion,the different strain of mice does not influence the results of dominant lethal test
