摘要
试验以苹果品种新红星、奥查金、美铃7为试材,对影响无菌繁殖系建立的两个主要因素即污染率和分化率进行了研究。结果表明:在4月下旬,各品种均已展叶4~5片时接种生长点,污染率较低,成活率较高,且易于生长分化。并发现不同品种间分化难易程度有很大差异,新红星是一个较难分化的品种,通过研究,其较适的组培条件是:改良C17+BA0.5mg/L+IBA0.2mg/L+CH300mg/L十蔗糖30g/L为起始培养基,接种后先光培养7d再暗培养7d,然后一直光培养直至分化。
The research was carried out in the rate of contamination and differentiation with three apple cultivars which played the main role in the inoculation of plantlet. From the results we knew that when inoculated in late April and bud sprout 4~5 leafs,it was easy to get lower contaminate rate,high survival rate, better growth and differentiation. Also there were great differences in differentiation among these cultivars. Starkrimson was difficult in differentiation. The better cultural condition for Starkrimson differentiation were as follows:first cultural medium with modification C17+BA 0. 5mg/L+IBA 0. 2mg/L+sucrose 30g/L, cultured it one week in the light and another week in the dark after inoculation,than in the light till it differentiated.
