摘要
目的探讨反义结缔组织生长因子(connective tissue grouth factro,CTGF)对兔耳增生性瘢痕的抑制作用。方法选择20只大耳白兔建立增生性瘢痕动物模型,随机分成5组,对照组为A组,注射转化生长因子β1(TGF-β1)为B组,注射CTGF为C组,先后注射TGF-β1和CTGF为D组,注射反义CTGF为E组,每个治疗组又分为治疗7、14、20d3组;通过逆转录-聚合酶链式反应(RT-PCR)方法检测增生性瘢痕中CTGF mRNA的表达,免疫组织化学检测CTGF蛋白表达,HE、Masson染色观察切片内成纤维细胞数密度,计算机辅助图像分析测算切片内胶原纤维面密度。结果E组中CTGF mRNA、蛋白表达、成纤维细胞数密度和胶原纤维面密度均降低,与A、B组比较,差异有统计学意义(P〈0.05)。结论反义CTGF可以抑制兔耳增生性瘢痕的增殖过程,使瘢痕组织纤维化程度明显减轻。
Objective To investigate the effect of antisense CTGF on inhibiting hypertrophic scar of rabbit ears. Methods 15 big ears white rabbits were used to establish the model of hypertrophic scar of rabbit ears, which was randomly divided into five groups. The hypertrophic scar were injected intralesionally with saline (group A), TGF-β1 (group B), CTGF (group C), TGF-β1, CTGF (group D), and antisense CTGF (group E), which were further assigned into 3 groups, i. e. , 7, 14 and 20 days after treatment. The expression of CTGF mRNA and protein was assessed by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry. The numerical density on area (NA) of fibroblasts was calculated with HE-stained sections and the area density on area (AA) of collagen was measured using a computer-assisted morphometrical system with Massonstained sections. Results The expression of CTGF mRNA and CTGF protein, NA of fibroblast and AA of collagen decreased in group E, which showed statistical difference compared with groups A and B. Conclusions The results suggest that antisense CTGF is able to inhibit the proliferation process of hypertrophic scar in rabbit ears and to remarkably decrease the degree of fibrosis in the scar.
