摘要
目的:探讨高原急性缺氧对大鼠食欲及其下丘脑胰升血糖素样肽-1 mRNA表达的影响。方法:雄性Wistar大鼠随机分为平原对照组、3000m缺氧组、4000m缺氧组、5000m缺氧组和6000m缺氧组,各缺氧组设置缺氧后1、2、3d3个时点。观察缺氧环境下大鼠食欲的变化;采用RT-PCR方法检测大鼠下丘脑中GLP-1 mRNA的表达。结果:①高原急性缺氧后,大鼠食欲减退。②对照组和缺氧组大鼠下丘脑中均存在GLP-1mR-NA的表达。高原急性缺氧后,下丘脑GLP-1 mRNA表达增加,并随着海拔高度的上升和缺氧时间的延长逐渐增加(P<0.05,P<0.01)。结论:高原急性缺氧后下丘脑GLP-1 mRNA表达增加,可能是大鼠食欲减退的重要原因之一。
AIM: To investigate the effect of acute hypoxia on the appetite and expression of glucagons like peptide- 1 (GLP- 1 ) mRNA in rat hypothalamus. METHODS: Wistar rats were randomly divided into control group, 3 000 m group, 4 000 m group, 5 000 m group and 6 000 m group for different times ( 1 d, 2 d and 3 d, respectively). The change of appetite was observed and the expression of GLP - 1 mRNA in hypothalamus of rats was measured using the technique of semi - quantitative reverse - transcription polymerase chain reaction. RESULTS : ① After acute exposure to hypoxia, the appetite of rats decreased. ② The expression of GLP - 1 mRNA in the test groups and the normal groups was detected in rat hypothalamus. After acute exposure to hypoxia, the expression of GLP - 1 mRNA was enhanced in rat hypothalamus with the increase in altitudes and time of acute hypoxia (P 〈0. 05, P 〈0. 01 ). CONCLUSION: Acute hypoxia might inhibit food intake in rats. After acute exposure to hypoxia, expression of GLP - 1 mRNA increases in rat hypothalamus, which may be one of the important reasons for loss of appetite.
