响应面法优化金线鱼胃蛋白酶提取条件的研究(英文) 被引量:2

OPTIMIZATION OF STOMACH PROTEASE PRODUCTION FROM NEMIPTERUS VIRGATUS(HOUTTUYN) BY RESPONSE SURFACE METHODOLOGY

摘要 研究了鱼体体重对金线鱼胃蛋白酶总活力的影响,并利用响应面法对金线鱼胃蛋白酶的提取条件(提取温度、提取时间和pH值)进行了优化.研究表明:在金线鱼的胃中,蛋白酶活力和种类随着鱼体重的增加而增加;响应面法优化获得的最佳提取条件为:提取温度23℃、提取pH5、提取时间10 h,提取的胃蛋白酶活力可达到1 881 U/g.min. The protease activity of stomach increased with increase of the body weight. Not only the activity but also the kinds of stomach proteases increased with the development of Nernipterus virgatus. High protease activity (1881 U/g · min) of a stomach protease from Nernipterus virgatus was obtained using a response surface methodology. The interaction of three variables, extract time, extract temperature and pH value, suggested to be insignificant variables. The model equation obtained was validated experimentally at optimum levels of the tested variables, extract time 10 h, extract temperature 23.22℃, pH 4.96. And its extract temperature and pH were adjusted to 23℃, pH 5 respectivly.
