黏蛋白MUC2、MUC5AC mRNA在分泌性中耳炎大鼠模型的表达 被引量:2

Expressions of MUC2 and MUC5AC mRNA in a Rat Model of Otitis Media with Effusion Induced by Ovalbumin

摘要 目的探讨变应性分泌性中耳炎分泌型黏蛋白MUC2、MUC5AC mRNA的表达变化及白三烯受体拮抗剂的抑制作用。方法建立卵白蛋白激发变态反应性中耳炎大鼠模型,实验动物分为对照组6只、实验组6只和干预组7只。采用RT-PCR技术分别检测MUC2、MUC5AC mRNA在3组中耳黏膜的表达变化。结果对照组中,MUC2 mRNA在中耳黏膜表达相对值为(0.23±0.08),MUC5AC mRNA则不表达;实验组表达相对值分别为(0.38±0.07)和(0.45±0.06),与对照组比较差别有统计学意义(P<0.01);干预组可部分抑制黏蛋白的表达,相对值分别为(0.28±0.07)和(0.29±0.09),与实验组比较差别有统计学意义(P<0.01)。结论MUC2、MUC5AC mRNA在变应性中耳炎中表达上调,可被白三烯受体拮抗剂部分抑制。 Objective To explore the expression level of mucins gene(MUC2,MUC5AC mRNA) in a rat model of otitis media with effusion induced by ovalbumin(OVA) and investigate the inhibitory effects of cysteinyl leukotriene receptor antagonist, montelukast, on the mucins output. Methods Nineteen Sprague-Dawley rats were randomly divided into three groups, treated, experimental and negative control. Rats from the experimental group were sensitized to OVA via intraperitoneal injection once a week for two weeks then challenged transbullarly with OVA 2 mg in PBS 50 μL. Middle ear mucosa was harvested and assayed for MUC2 and MUC5AC mRNA expressions by semi-quantitative reverse transcription-polymerase chain reaction. Results MUC2 and MUCSAC mRNA expressions were (0.23±0.08) in middle ear mucosa of the negative control and significantly up-regulation were observed after OVA chal lenge[(0.38±0.07) vs (0.45±0.06), (P〈0.01)]. Montelukast could partially inhibit the expression of MUC2 and MUC5AC mRNA[(0.28±0.07) vs (0.29±0.09), (P〈0.01)]. Oonclusion The expression level of MUC2 and MUC5AC mRNA significantly up-regulated in OVA-induced otitis media and could be down-regulated by cysteinyl leukotriene receptor antagonist.
