摘要
建立了高效液相色谱-电化学检测法测定美沙拉秦中有关物质3-氨基水杨酸、2-氨基苯酚和4-氨基苯酚。以辛烷基硅烷键合硅胶为填充剂,以磷酸二氢钾-辛烷磺酸钠缓冲溶液(pH2.2)-甲醇-乙腈(890∶80∶30)为流动相A,甲醇为流动相B进行梯度洗脱,以玻碳圆盘电极为工作电极,银-氯化银电极为参比电极,于+0.7V电压处测定。美沙拉秦及其3个有关物质的线性范围分别为0.2~1.8、0.2~1.8、0.02~0.18、0.02~0.2μg/ml,检测限均为0.02ng。
A HPLC method with electrochemical detection was established for the determination of the related substances-3-aminosalicylic acid, 2-aminophenol, 4-aminophenol, in mesalazine. A C8 column was used, with the mobile phase of potassium dihydrogen phosphate-sodium 1-octanesulfonate buffer solution (pH 2.2)-methanol-acetonitrile (890 : 80 : 30) and methanol by gradient elution, at 0.7V. The glassy carbon electrode and silver-silver chloride electrode were used as the working electrode and reference electrode, respectively. The calibration curves of mesalazine and its related substances were linear in the concentration range of 0.2 - 1.8, 0.2 - 1.8, 0.02 - 0.18 and 0.02 - 0.2μg/ml, respectively. The limit detection of four components was 0.02ng.
