摘要 目的研究2,3-吲哚醌(2,3-indolinedione,inatin,ISA)的抗衰老作用及其机制。方法昆明种小鼠随机分为6组:空白对照组,模型组,卵磷脂组(阳性对照组),ISA低、中、高剂量组。用Y型电迷宫法,测定小鼠的学习记忆能力;测定胸腺指数、脾指数和脑指数;测定脑组织中超氧化物歧化酶(SOD)活性,活性氧(ROS)和脂褐素(LF)含量。结果ISA中、高剂量组和卵磷脂组学习次数[分别为(14.33±4.32)次和(16.33±5.36)次]低于模型组(21.71±7.04)次(P〈0.05);反应正确率[分别为(90.00±11.55)%和(87.14±11.38)%]高于模型组(69.29±12.72)%,P〈0.05);胸腺指数、脾指数和脑指数明显增高(P〈0.01);与模型组比较,ISA提高SOD活性[(65.64±14.49)μmg^-1 protein vs(50.23±15.88)μmg^-1 protein,P〈0.05],降低ROS[(442.59±97.92)μmg^-1 proteinvs(540.20±89.02)μmg^-1 protein]和LF含量[(0.96±0.26)μmg^-1 proteinvs(1.31±0.23)μmg^-1 protein,P〈0.01]。结论ISA具有抗衰老作用,与其增强免疫功能和抗氧化有关。 Objective To investigate the anti-aging effects and mechanism of 2,3-indolinedione (isatin, ISA). Methods Kunming mice were divided into 6 groups randomly: control group, model group, lecithin group, and 3 ISA treatment groups (50 mg · kg^-1 , 100 mg · kg^-1 , 200 mg · kg^-1 ). Every mouse was administered once a day for a continuous 40-day period. The Y-maze test was operated to test the ability of learning. Followingly, the thymus gland, spleen and brain tissue were carefully removed and separately weighed. And the contents of SOD, ROS and LF in brain tissues were assayed. Results Lecithin and ISA exhibited increase in learning and memory abilities ( P 〈 0.05 ), raised the brain, spleen and thymus indexes ( P 〈 0.01 ) , increased the activity of SOD (65.64 ± 14.49 vs 50.23 ± 15.88, P 〈 0.05 ) in the brain ( P 〈 0.01 ), decreased the level of ROS (442. 59 ±97. 92 vs 540. 20 ± 89. 02) and LF(0. 96 ±0. 26 vs 1. 31 ±0. 23) (P 〈0. 01). Conclusion ISA exerts anti-aging effects via increasing immunity and anti-oxygenation.
