摘要
目的探讨DEHP对的小鼠的遗传毒性及氧化损伤。方法用不同剂量的邻苯二甲酸二异辛酯[di-(2-ethyl-hexyl)phthalate,DEHP]对小鼠每日腹腔注射染毒2周,通过彗星实验检测染毒2周小鼠的脑细胞DNA损伤,并测定肝,睾丸,脑组织中超氧化物歧化酶的活性变化。结果随着DEHP浓度增高,小鼠脑组织细胞DNA的损伤随之加重,DEHP的浓度达到125mg/kg和375mg/kg时,其可造成DNA的显著断裂(P<0.01)。肝、睾丸、脑的SOD活性变化是随DEHP浓度由低到高而先升高后降低,375mg/kg与对照组有显著性差异。结论DEHP可造成小鼠脑组织细胞DNA的损伤,及各脏器的氧化损伤。
Objective To study genotoxicity and oxidative damage induced by di -ethylhexyl phthalate(DEHP) in mice everyday. Methods Different dose of di - ethylhexyl phthalate were injected at the abdomen of the mice for 2 weeks. Examine the effect of di - ethylhexyl phthalate(DEHP) inducing DNA damage in nerve system of mice by celiac injection with single cell gel electrophoresis(SCGE) and the changes of the activity of superoxide dismutase (SOD) in liver, testicle and brain. Results DEHP can induce DNA damage of nervy system of mice and DNA damage rises along with concentration of DEHP( 125,375,500 mg/kg rising. At the concentration of 125 mg/kg and 375 mg/kg ,DEHP can induce DNA strand breaks(P 〈 0.01 ). The activity of superoxide dismutase (SOD) in liver, testicle and brain first rise and then decrease alone with the dose. There are significant different in 375mg/kg group compared with that of the control. Conclusions DEHP can induce DNA damage in nerve system of mice and oxidative damage in internal organs of mice ,existing evident doseage - effect connection.
