摘要
目的观察大黄素对HSC-T6细胞凋亡及胞内钙离子浓度的的影响。方法将培养细胞分成正常组和大黄素不同浓度干预组(大黄素终浓度分别为10μmol/L、20μmol/L、40μmol/L),采用流式细胞仪观察各组细胞凋亡率;采用激光共聚焦显微镜检测胞内钙离子浓度的变化。结果不同浓度大黄素诱导HSC-T6的凋亡率分别为4.0±0.5%、4.3±0.6%、6.9±1.1%,与正常组比较有显著性差异(P<0.05);不同浓度大黄素导致HSC-T6细胞胞内钙离子浓度明显增高,其变化率分别为102.50%、125.10%、215.00%,与对照组比较有显著性差异(P<0.01)。结论大黄素有诱导HSC-T6细胞凋亡的作用,其机制可能与引起HSC-T6钙超载有关。
Objective To explore the effect of emodin on the apoptisis and intracellular Ca^2+ concentration of Hepatic Stellate Cell - T6. Methods The immortalized rat hepatic stellate cell line HSC - T6 was commonly allocated into normal control group and treated groups with different concentrations emodin(the extreme concentration are 10μmol/ml, 20μmol/ml, 40μmol/ml respectively). The cell apoptosis rate were analyzed by flow cytometry, and intracellular Ca^2+ concentration of HSC -T6 were detected by Furo - 3/Am fluorescence assay and confocal laser microscopy. Results After intervention of different concentration emodin,the apoptosis rate of HSC - T6 is 4.0 ± 0.5% , 4.3 ± 0.6% , 6.9 ± 1.1% respectively, and there are statistical significance compared with the control group(P 〈 0.0.5 ). Intracellular Ca^2+ concentration of HSC - T6 were significantly increased by incubation with Emodin . The vary ratio of Ca^2+ is 102.50% , 125.10% ,215.00% respectively, and obviously difference appear compared with the control group(P 〈 0.01 ). Conclusions Emodin can induce the apoptosis of HSC -T6,and its prossiblly mechanism is to increase the intracellular Ca^2+ concentration of HSC -T6.
