摘要
[目的]探讨细胞色素P450(cytochromeP450,CYP450)基因多态性与重度牙周炎易感性关系。[方法]采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术,对河南地区66例重度牙周炎病例和40例正常对照的CYP2E1基因型进行检测。[结果]在重度牙周炎病例组和对照组中,RsaⅠ识别的CYP2E1野生型(C1/C1)基因频率分别为59.1%和60.0%(P﹥0.05);DraⅠ识别的CYP2E1野生型(DD)基因频率分别为51.5%和37.5%,MspⅠ识别的CYP1A1突变纯合型(m2/m2)基因频率分别为27.3%和12.5%,差异均无统计学意义(OR值分别为0.963、1.174和0.381;P分别为0.926、0.689和0.0737)。[结论]CYP2E1基因RsaⅠ、DraⅠ多态性和CYP1A1基因MspⅠ多态性与河南地区重度牙周炎易感性可能无关。
[Objective] To investigate the relationship between CYP450 gene polymorphisms and the susceptibility of severe periodontitis. [Methods] Buccal swabs from 66 patients with severe periodontitis and 40 periodontal healthy con.trois were obtained. DNA was extracted from each sample using Chelex-100 method, then PCR-RFLP was performed to determine the CYP450 genotype. Data were analyzed with statistics software. [ Results] The results showed that in patients with severe periodontitis and cases in the control group, the frequency of the wild-type (C1/C1) detected by Rsa [ digestion was 59.1% and 60% respectively, the frequency of wild-type (DD) detected by Dra I digestion was 51.5% and 37.5% respective, the fre- quency of variant-type (m2/m2) detected by Msp I digestion was 27.3% and 12.5% respectively. No statistical difference of genotype distribution was found between the patients and cases in the control group (OR= 0.963, 1.174 and 0.381 respectively. P = 0.926, 0.689 and 0.0737 respectively) . [Conclusion] No direct relationship exists between CYP450 gene pulymorphisms and the risk of severe periodontitis.
