摘要
[目的]探讨青链霉素+庆大霉素防控细胞培养中枯草杆菌污染的可行性。[方法]采用不同浓度青链霉素和庆大霉素配伍的方法,观察其抑制枯草杆菌的效果及对STO转基因小鼠滋养层细胞和D3系小鼠胚胎干细胞的生长情况的影响。[结果]700u/ml青链霉素和280u/ml庆大霉素联合应用的抑菌效果最佳,且对小鼠胚胎干细胞和小鼠成纤维细胞的生存率无明显影响。[结论]采用青链霉素和庆大霉素可以有效控制细胞培养中枯草杆菌的污染。
[Objective] To study the feasibility of preventing hay bacillus contamination in cell culture by nrycillin and gentamyein. [ Methods] Myllicin and gentamycin were added into medium with different concentration, the survival of hay baciLlus, mice embryonic stem cells, and mice was observed. [Results] The combination of 700 u/ml nrylicin and 280 u/ml gentamycin was better than others in preventing hay bacillus, and it did not obviously affect the survival of mice embryonic stem cells, as well as fibroblast. [Conclusion] Mycillin and gentamycin can be used to prevent hay bacillus contamination in cell euhure.
