β_2-AR基因+491C/T多态性与新疆哈萨克族男性原发性高血压的关联研究 被引量:3

摘要 目的:研究陡肾上腺素能受体(β2-AR)基因+491C/F多态性与新疆男性哈萨克族人原发性高血压(Essential Hypertension EH)的关系。方法:以人群为基础的病例一对照研究,选择175例30岁.60岁男性哈萨克族人(83例EH者与92例血压正常者)为研究对象,酚/氯仿法提取外周血白细胞基因组DNA,应用PCR—RFLP技术,检测+491C/T基因型,并观察各基因型、等位基因频率在该人群中分布及其差异。结果:只检测出两种基因型CC、CT,分布频率为9r7.71%、2.29%,等位基因C,T频率为98.86%、1.14%。CC、CT在EH组分布频率分别为95.18%、4.82%,正常血压组中未检出CT基因型。经Fisher’s精确概率检验,两组基因型、等位基因分布频率差异均有统计学意义;基因型间收缩压差异有统计学意义。结论:β2-AR基因+491C/T多态性与新疆男性哈萨克族人EH尤其是高收缩压相关联,提示该多态性可能是男性哈萨克族人EH的易感因素。 Objective: To study the molecular pathogenic mechanism of predispose to hypertension in Kazakns, we investigated the association between the + 491C/T polymorphism of β2- AR gene and essential hypertension in Kazakns of Xinjiang in male. Methods:In this population- based ease- control study, we recruited 175 Kazakns subjects (83 hypertensives and 92 normotensives ,30- 60 years old) from the pastoral areas in Xinjiang . Peripheral blood were collected from all recruited subjects. The DNA was extracted by means of a standant phenol- chloroform method. Then the genotypes of β2 - All was identified by PCR - restriction fragment length polymorphism in all the subjects. The distribution of genotypic and allelic frequencies between hypertensives and normotensives were studied. Results: In the population, the frequencies of the genotype CC, CT were 97.71% and2.29% respectively, no IT was found, the genotypic and allelic frequencies were in Hardy - Weinberg equilibrium(Χ^2 = 0.023, P = 0. 878). These two genotypic frequencies were 95.18% and 4.82% respectively in essential hypertensives, while no CT in normotensives. The allelic frequencies of C,T were 97.59% ,2.41% and 100% ,0% in two groups and there is a significant differences between these two groupso In addition, systolic blood pressure has significant difference in the groups classified according to genotypes. Conclusion: The + 491C/T polymorphism of β2 - All gene may be associated with essential hypertension in male Kazakns, especially in systolic blood pressure . which implicates that it is possible one of the susceptible factors of essential hypertension in Kazakns of Xinjiang.
