mTOR信号通路调控HK-Ⅱ表达对结肠癌细胞增殖的影响 被引量:5

摘要 目的:探讨mTOR信号通路是否可通过调控HK-II蛋白表达的途径来影响结肠癌细胞增殖。方法:Western印迹检测结肠癌细胞mTOR、HK-Ⅱ蛋白表达水平,并观察抑制mTOR信号通路对HK-Ⅱ蛋白表达的影响。采用MTT法检测mTOR和HK-Ⅱ特异性抑制剂作用后结肠癌细胞增殖的变化。结果:mTOR、HK-Ⅱ蛋白在4种结肠癌细胞中均明显表达;抑制mTOR可阻断结肠癌细胞HK-Ⅱ蛋白表达;抑制mTOR和抑制HK-Ⅱ均可明显控制结肠癌细胞增殖,且两者有协同效应。结论:mTOR信号通路可能通过调控HK-Ⅱ的表达影响结肠癌细胞增殖。 Objective: To analyze if mTOR signaling pathway might affect colon cancer cell proliferation through regulating expression of HK - Ⅱ protein. Methods : Expressions of mTOR and HK - Ⅱ were examined by Western blot test, and inhibition of roTOR on HK - Ⅱ protein expression was also detected. After colon cancer cell lines were incubated with mTOR specific inhibitor and HK -Ⅱ specific inhibitor respectively, cell growths were assessed using MTT assay. Results. High expressions of mTOR protein and HK - Ⅱ protein were observed in four kinds of colon cancer cell lines, mTOR specific inhibitor suppressed expression of HK - Ⅱ protein. Inhibition of mTOR and HK - Ⅱ both significantly suppressed colon cell growth. Moreover, combination of mTOR inhibition and HK - Ⅱ inhibition produced synergistic cytotoxic effect. Conclusion: mTOR signaling pathway may affect colon cancer cell proliferation through regulating expression of HK - Ⅱ protein.
