长期吸入性烟尘对大鼠肺组织超微结构及TGF-β_1表达的影响

摘要 目的观察长期吸烟对肺组织超微结构和TGF-β1表达的影响,探讨TGF-β1在长期吸烟所致肺病理变化中的作用。方法建立Wistar大鼠吸烟模型,电镜下观察吸烟对大鼠肺组织超微结构的影响,应用免疫组织化学法检测TGF-β1在肺组织中的表达。结果与对照组比较,随着吸烟时间的延长,吸烟组大鼠肺泡细胞细胞器出现损伤并逐渐加重,5个月最重;随着吸烟月份的增加,吸烟组肺组织TGF-β表达增多,至5个月时表达最多,6个月表达减少。结论吸烟可导致肺组织细胞超微结构破坏和TGF-β1表达增强,说明肺组织破坏与TGF-β1表达有相关性。 Objective To observe the effect of long-term smoking on the ultrastructure of lung cells and expressions of TGF-β1 in rat lung tissues .To study the role of TGF-β1 on smoking-induced lung pathological changes. Methods To establish the model of Wistar rats smoking. The changes of ultrastructure of lung cells were detected by electron microscope. The expressions of TGF-β1 and TGF-β1 mRNA in lung tissues were observed by immunohistochemistry. Results Compared with the control groups, there were some damages of the organelles of alveolus cells from the smoking groups under the electron microscope. Along with the smoking time, the lung damages aggravates and it was the most conspicuous in the fifth month smoking group. The expressions of TGF-β1 and TGF-β1 mRNA in lung tissues increase gradually among smoking months. It was the most conspicuous in the fifth month. Conclusion Smoking may result in the damages of the ultrastructure of alveolus celIs, and along with the smoking months, the damages aggravates. The changes may correlate with the expressions of TGF-β1.
