摘要
通过添加不同浓度的牛卵泡液(bFF),探讨了bFF对牛卵母细胞体外核成熟率的影响;并筛选出bTCM-199(10%)培养体系与TCM-199培养体系进行核成熟效果的对比试验。结果表明:20%、30%bFF组处在GV期和MⅠ期卵母细胞比率极显著高于10%组(P<0.01),而卵母细胞核成熟(处于MⅡ期)率10%组显著高于20%和30%组(P<0.01);TCM-199培养体系与bTCM-199(10%)培养体系2组减数分裂各阶段卵母细胞比率基本一致,前者卵母细胞核成熟(处于MⅡ期)率与后者差异不显著(P>0.05)。与bTCM-199(10%)培养体系相比,TCM-199培养体系成分简单,是理想的牛卵母细胞体外成熟培养体系。
Effect of bFF on the maturation rate of bovine oocyte nucleus in vitro was studied by adding bFF with different concentration. The nuclear maturation effectiveness in TCM -199 culture system and bTCM-199 (10 %) culture system was compared in this paper. The results indicated that the ratio of oocytes in GV and M Ⅰ stage in group with 20 % and 30 % bFF was extremely higher than that of group 10 % (P〈0.01), while the nuclear maturation (M Ⅱ ) rate of group 10 % was extremely higher than that of group 20 % and 30 % (P〈0.01). The ratio of oocytes in different meiosis stages in TCM-199 culture system was the same as that in bTCM-199 (10 %) culture system. And there were no differences in the nuclear maturation (M Ⅱ ) rate between the former and the latter (P〉0.05). Compared with bTCM -199(10 %) culture system, the composition was simple in TCM-199 culture system, which was an ideal culture system for in vitro maturation of bovine oocyte.
