摘要 目的:观察慢性乙醇摄入对大鼠单个肝窦内皮细胞(LSEC)结构的影响。方法:采用乙醇直接灌胃的方法建立酒精性肝病大鼠动物模型,应用稍加改良的Pertoft梯度离心方法对不同阶段的LSEC进行分离纯化,用透射电镜观察不同时期LSEC形态和结构的改变。结果:乙醇灌胃4周末、8周末、12周末LSEC在形态、窗孔大小和数量上均出现不同程度的改变;停止乙醇灌胃12周末,大部分细胞比乙醇灌胃12周末时有明显的恢复。结论:乙醇的慢性刺激使细胞形态改变;停止刺激后,细胞的形态、窗孔大小和数量都有一定的恢复。 Objective: To observe the effect of chronic ethanol intake on the structure of liver sinusoidal epithelial cell ( LESC ) in rats. Methods: The rat model of alcoholic liver disease was established by gastric administration of ethanol. The LESCs of different stages were isolated and purified with modified Pertoft gradient centrifuge, and the morphological changes of LESCs were observed under transmission electron microscopy. Results: At the end of week 4, 8, and 12 of ethanol administration, there were changes in cell shape, sinusoidal fenestrae size, and cell number at various degrees. At the end of week 12 after stopping ethanol administration, most cells recovered significantly. Conclusion : The morphology of LSECs changes after chronic stimulation of ethanol and recovers to some extent after ethanol stimulation stops.
