摘要
目的:探讨三氧化二砷(As2O3)对卵巢癌细胞HO-8910体外生长的影响及其相关机制.方法:用倒置显微镜对细胞进行观察,用免疫细胞化学染色法检测p53基因表达.结果:(1)三氧化二砷作用浓度在0~12.0 μmol/L时,随着剂量的增加对卵巢癌细胞抑制作用增强.(2)卵巢癌细胞p53基因表达增强.结论:较高浓度三氧化二砷对卵巢癌细胞株HO-8910细胞有生长抑制作用并呈剂量-时间依赖效应 As2O3作用引起卵巢癌细胞HO-8910细胞凋亡的机制与p53基因表达增强有关.
Objective:The study was to investigate the effects of arsenic trioxide on the proliferation of ovarian cancer cell line HO - 8910 in vitro and relative mechanism. Methods: Morphological changes of cells were observed. Immunocytochemical staining was used to examine the expression of p53 gene. Results: ( 1 ) In vitro, arsenic trioxide of 0 -12.0μmol/L showed significant cor -relation to the HO -8910 cellular proliferation , and the degree of the inhibition enhanced with the increasing dose. (2) Higher concentrations of arsenic trioxide up - regulated p53 gene expressions was intensive. Conclusion:Arsenic trioxide at high concentrations could obviously inhibit the proliferation of ovarian cancer cell line HO- 8910 in vitro, and there was an dose- response relationship; the apoptosis mechanism of ovarian cancer cell line HO - 8910 was related with up - regulating of p53 gene expression.
