蒿甲醚用于卡氏肺孢子虫肺炎大鼠的治疗及其对IL-6影响的研究 被引量:4

摘要 目的从病理学和细胞因子角度探讨蒿甲醚对大鼠卡氏肺孢子虫肺炎(Pneumocystis carinii PneumoniaPCP)的治疗效果和作用机理。方法给SD大鼠皮下注射地塞米松磷酸钠建立卡氏肺孢子虫肺炎动物模型,治疗组给予蒿甲醚治疗。ELISA双抗夹心法分别检测血清和支气管肺泡灌洗液中IL-6水平。结果与感染对照组比较,蒿甲醚治疗组症状显著改善、肺印片中卡氏肺孢子虫包囊数目显著减少、肺组织炎症明显减轻、血清和肺泡灌洗液中IL-6水平明显下降。结论蒿甲醚具有一定抗大鼠卡氏肺孢子虫肺炎作用,能够降低PCP大鼠IL-6。 To observe the therapeutic effect of Artemether (ATM) on P. carinii pneumonia model of rats and its mechanism of action from point of view of pathology and cytokine production, the infection model of Sprague-Dawley (SD) rats was established by subcutaneous injection of dexamethasone sodium phosphate, in which ATM was given in the experimental group of rats and the level of IL-6 in sera and broncheoalveolar lavage fluids (BALFs) was determined by double-antibody sandwich ELISA assay. It was demonstrated that compared with the infection control group, the symptoms of rats occurring in the ATM treatment group were greatly relieved. Also, the number of Pneumocystis carinii cysts was markedly reduced in the lung print smears and the inflammatory changes in the lung tissues were obviously lightened in rats treated with ATM. In addition, the level of IL-6 in sera and BALFs were obviously decreased with significant difference with that of the infection control group. It is concluded that ATM is effective for the treatment of P. carinii pneumonia and it can reduce the production of IL-6 in rats.
