磷制剂对砷染毒细胞增殖抑制作用影响的实验研究 被引量:2

摘要 目的探讨磷制剂对NaAsO2染毒的Vero细胞体外增殖抑制的影响。方法用不同浓度的NaAsO2处理细胞后,在培养液中加入不同剂量的磷制剂,通过噻唑蓝还原法(MTT)观察对细胞增殖抑制的影响,并用维生素E作为标准对照。结果一定剂量NaAsO2对Vero细胞增殖有抑制作用;207.00 mg/L,103.50 mg/L,51.75 mg/L三种剂量的磷酸钠及53.3 mg/L,26.65 mg/L的卵磷脂对砷染毒的细胞具有保护作用,可拮抗砷对细胞的增殖抑制作用,与维生素E的拮抗作用相比,高剂量的磷酸钠与维生素E相当,中、低剂量的磷酸钠及高、中剂量的卵磷脂的保护作用次于维生素E,而次磷酸钠的作用相反,与砷的毒性具有协同作用。结论磷制剂对对砷染毒细胞增殖抑制作用是双向的,与磷的不同价态、不同化合物在体内代谢不同有关。 Objective To discuss the influence of trisodium phosphate, sodium hypophosphite and lecithin on multiplication - inhibition of vero cell in vitro contaminated by arsenic. Methods After processed ceil by different density of NaAsO2 , different dosages of trisodium phosphate, sodium hypophosphite and lecithin were added to nutrient fluid, then the influence of MTT assays on ceil multiplication - inhibition was observed with the comparison of vitamin E with the standard control. Results Certain dosages of NaAsO2 had the inhibitory action to the vero ceil multiplication; Three kinds of dosage(207.00 mg/L, 103.50 mg/L, 51.75 mg/L) of trisodium phosphate and two kinds of dosage of lecithin(53.3 mg/L,26.65 mg/L) had the protective function to the cell contaminated by arsenic, that could rival the arsenic in cell multiplication - inhibition. Compared with vitamin E, the antagonistic effect of high dosage trisodium phosphate was equal, while the effect of middle, low dosage was poor. The protective function of high, middle dosage of lecithin was inferior to vitamin E, but the function of sodium hypophosphite was opposite. Conclusions The effect of trisodium phosphate, sodium hypophosphite and lecithin on multiplication - inhibition of ceils induced by arsenic is bidirectional. It is relative to the different states of phosphorus and the metabolism in vivo of different compounds.
