摘要 目的:检测肿瘤坏死因子α(TNF-α)在大鼠脑外伤后急性胃黏膜病变前后的含量并探讨其意义.方法:采用改良的Allen法建立大鼠颅脑外伤并发急性胃黏膜病变模型,同时设立假手术对照组,测定不同时间点(1,3,6,24h)各组大鼠血清中TNF-α含量及胃黏膜溃疡指数,并观察胃黏膜大体及光镜下组织病理学改变.结果:模型组1,3,6,24h血清中TNF-α含量明显高于对照组(t=16.45,P=0.000;r=5.252,P=0.000;t=9.099,P=0.000;t=12.028,P= 0.000).模型组胃黏膜损伤严重,6,24h点溃疡指数明显高于对照组(t=7.275,P=0.000;t= 10.579,P=0.000),TNF-α含量与溃疡指数呈线性正相关(r=0.68,P=0.004),组织学也有相应改变.结论:TNF-α在颅脑外伤急性胃黏膜病变的发病中起一定作用. AIM: To detect the content of tumor necrosis factor-α (TNF-α) in acute gastric mucosal injury in rats after acute brain trauma and explore its significance. METHODS: Modified Allen's method was used to induce the rat model of acute gastric mucosal injury after acute brain trauma. Meanwhile, the sham operation group was also designed. The content of TNF-α and ulcer index of gastric mucosa were determined at different time points (1, 3, 6, 24 h), and the pathological changes were also observed under light microscope. RESULTS: The content of TNF-α in model group was significantly higher than that in control group at the Ist, 3^rd, 6^th and 24^th hour (t = 16.45, P = 0.000; t = 5.252, P = 0.000; t = 9.099, P = 0.000; t = 12.028, P = 0,(300). More severe injury occurred in model group and the ulcer index of gastric mucosa was markedly higher than that in control group at the 6th and 24th hour (t = 7.275, P = 0.000; t = 10.579, P = 0.000). TNF-α ontent was positively correlated with the ulcer index of gastric mucosa (r= 0.68, P = 0.004). CONCLUSION: TNF-α plays a certen role in acute gastric mucosal injury after acute brain trauma.
